Abstract:

Aimed to develop a protocol for in vitro propagation of Gabirobeira from nodal segments and to characterize anatomically and ultrastructurally the calogenesis. In the first experiment, shoots were induced in vitro using WPM (Wood Plant Medium) culture medium with different concentrations of 6-benzylaminopurine (0, 0.25, 0.5, 1.0 and 2.0 mg L- 1), in the presence or absence of photosynthetically active radiation. In the second experiment, rhizogenesis was induced using different concentrations (0, 0.5, 1.0, 1.5 and 2.0 mg L-1) of indolbutyric acid (IBA), indolacetic acid (IAA) and naphthalenoacetic acid (ANA). The statistical design was completely randomized with 5 repetitions (each repetition composed of 3 tubes) in both experiments, being for the shoots 2x5 factorial scheme (two environments and five concentrations). In the third experiment, calli were induced in nodal segments using different concentrations (0, 1.0, 2.0 and 4.0 mg L-1) of IBA, IAA, ANA, 2,4-dichlorophenoxyacetic and Picloram. The indicated concentration for shoot induction is 1.0 mg L-1 BAP kept in the dark for elongation to occur. With the tested treatments it wasn't possible to obtain efficient protocol for rhizogenesis. Callus was induced at all concentrations tested except the control, but 1.0 mg L-1 picloram was more efficient. Through the callus growth curve together with the histochemical and ultrastructural analyzes, it is possible to infer that on the 28th day the calluses must be transferred to new culture media, since during this period it's possible to visualize the meristematic center formation standing out from the periphery.
