Abstract:

Guanandi (Calophyllum brasiliense Cambess), is a Brazilian native species of economic forestry and medicinal value. The aim of this work was to improve the in vitro germination, multiplication and elongation of Guanandi. In the germination experiment the seeds were osmoconditioned with potassium nitrate solutions (KNO3) at 0%, 0.2%, 0.5%, 1%, 2%, 5%, 10%, 15%, 20% 25% and 50% or inoculated in WPM (Wood Plant Medium), ½WPM, MS, ½MS and H2O. In the multiplication experiment the nodal segments were inoculated in WPM culture medium supplemented with citokins: 6- belzilaminopurina (BAP) (2; 4,4; 8:16 µM), kinetin (CIN) (4, 6, 8 and 16 µM), 6th -isopentyl-amino purine (2-iP) (8 and 16 µM) and thidiazuron (TDZ) (0.91, 2.27, 4, 8 and 16 µM) and 0 µM as control. In the elongation experiment the explants were inoculated on WPM medium supplemented with gibelelic acid GA3 (0, 5, 10 and 20 uM) and manteined on dark or 16h foteoperiod. Germination with potassium nitrate (KNO 3) we observed that there is a correlation between the germination percentage and the percentage of KNO 3, showing that at 30, 45 and 60 days linear regression equation explains the dependent variable (Y). The percentage of germination and seed vigor decreased with respect to the concentration of KNO3. Treatment 50% was toxic impeding the germination of seeds. Regarding the different types of media, there was no significant effect of culture media for the germination percentage at 15 and 30 days and the emergency speed index. The multiplication of treatment showed no significant difference in the average size of the shoots, average number of buds and number of leaves at 30 and 60 days. With regard to different concentrations of GA3, in different lighting conditions the explants kept in light showed higher average size of buds, the greater number of shoots and leaves and smaller oxidation rate compared to those kept in the dark.
